What Does Water Sampling Test For? A Guide to Legionella, Pseudomonas, TVC, E. coli & Coliforms

Water sampling can provide valuable information about the microbiological condition of a water system, but not every water test looks for the same thing.


Depending on the property, water system and reason for investigation, testing may include Legionella, Pseudomonas aeruginosa, Total Viable Counts (TVC), E. coli and coliform bacteria.


Understanding what each test measures and, equally importantly, what it does not tell you— can help property managers, facilities teams, landlords and other duty-holders make informed decisions about water hygiene and risk management.


Why Is Water Sampling Carried Out?


Water sampling may be appropriate for a number of reasons, including:


  • Investigating concerns about the microbiological condition of a water system

  • Supporting an established water hygiene monitoring programme

  • Investigating unexpected changes or suspected contamination

  • Assessing particular outlets or parts of a water system

  • Supporting corrective or remedial works

  • Checking conditions following intervention where appropriate

  • Monitoring higher-risk or specialist environments


The appropriate test, sampling locations and frequency should be determined by the water system, its use, the people who may be exposed and the reason for sampling.


Water sampling is not a substitute for appropriate risk assessment or effective management of the water system.

What Does Legionella Water Sampling Test For?


Legionella testing looks for Legionella bacteria within a water sample, with laboratory results typically reported as colony-forming units per litre (CFU/L).


Legionella bacteria can proliferate where water-system conditions allow, particularly where temperatures, stagnation, nutrients or system design provide favourable conditions.


Importantly, routine Legionella sampling is not required for every hot and cold water system.


HSE guidance states that microbiological monitoring of ordinary hot and cold water systems is generally not routinely required. Sampling may, however, be appropriate where there is doubt about the effectiveness of the control regime or where recommended temperatures, disinfectant concentrations or other precautions are not being consistently achieved.


Where Legionella monitoring is appropriate, HSE advises that samples should be tested by a UKAS-accredited laboratory participating in an appropriate water microbiology proficiency-testing scheme.


What is Pseudomonas Aeruginosa?


Pseudomonas aeruginosa is an environmental bacterium that can occur in water and wet environments.


It is an opportunistic pathogen and is particularly significant for people who are more susceptible to infection. UK guidance gives particular attention to its management in healthcare and augmented-care environments.


Water outlets, associated components and biofilms can provide environments in which the organism may persist.


When might Pseudomonas sampling be relevant?


Its significance depends heavily on the setting.

Testing can be particularly important within appropriate healthcare and other higher-risk environments, where water-system management and sampling strategies should be based on the relevant guidance and risk assessment.


It should not simply be treated as another routine test required for every property.


What Does TVC Mean?


TVC stands for Total Viable Count.

Unlike a Legionella or E. coli test, TVC does not normally identify one particular bacterial species.


Instead, laboratory culture is used to provide an indication of the number of viable microorganisms capable of growing under the specified test conditions.


TVC testing is commonly performed using different incubation temperatures, including 22°C and 37°C.


Broadly, these results can help provide information about the general microbiological condition of the water and can be particularly useful when looking for changes or trends over time.


Does a high TVC mean the water is dangerous?


Not necessarily.


A TVC result should not be interpreted as a direct count of harmful bacteria or as a simple standalone measure of whether water is “safe” or “unsafe”.


Its significance depends on the water system, sampling point, historical results, test conditions and reason for sampling.


A change from the system's normal pattern may sometimes be more informative than viewing a single result in isolation.


This is why laboratory results need appropriate interpretation rather than simply comparing every bacterial count against a universal number.


What Does E. coli Testing Tell Us?


Escherichia coli (E. coli) is used as an important microbiological indicator of faecal contamination.


For drinking water in England and Wales, the Drinking Water Inspectorate specifies a value of 0 E. coli per 100 ml at consumers' taps.


Detection of E. coli therefore warrants appropriate investigation.


The response will depend on the type of water supply, location, circumstances and applicable standards.


Testing for E. coli is particularly relevant where there are concerns about drinking-water quality, private water supplies, storage or potential contamination.


What Are Coliform Bacteria?


Coliforms are a broader group of bacteria used as indicators in water-quality assessment.

Unlike E. coli, the presence of a coliform organism does not automatically mean that recent faecal contamination has occurred.


However, unexpected coliform detection can indicate that further investigation of the water supply, treatment, storage or distribution system may be required.


For this reason, E. coli and coliform results should not be treated as interchangeable.


The applicable standards and interpretation also depend upon the type of water supply and the point at which the sample was collected.


Why Does the Sampling Location Matter?


A laboratory can only analyse the water that reaches it.


Choosing where to collect samples is therefore an important part of a meaningful sampling strategy.


Depending on the investigation, sampling points might include:


  • Representative hot and cold water outlets

  • Sentinel outlets

  • Little-used outlets

  • Water storage

  • Areas where control parameters have been difficult to maintain

  • Locations associated with a particular concern

  • Pre- and post-intervention locations where appropriate


For Legionella monitoring, HSE specifically emphasises the importance of selecting representative outlets and interpreting results in the context of the system.

Water Sampling vs Legionella Risk Assessment: What's the Difference?


The two should not be confused.


A Legionella Risk Assessment (LRA) considers the water system and identifies factors that could create or increase the risk of Legionella exposure.


This can include system design, temperatures, water storage, outlets, areas of stagnation, vulnerable occupants and existing control measures.


Water sampling, on the other hand, provides laboratory information about the particular water collected from a particular sampling location at a particular time.

HSE places risk assessment at the heart of Legionella control. Duty holders are required to identify and assess sources of risk and, where risks exist, implement appropriate measures to prevent or control them.


A water sample therefore does not replace a Legionella Risk Assessment.


Likewise, HSE makes clear that landlords are not required to obtain a “Legionella test certificate”.


What Happens if a Water Sample Produces an Unexpected Result?


An unexpected result should be considered in context rather than viewed in isolation.

Review → Investigate → Assess Controls → Take Appropriate Corrective Action → Re-sample Where Required → Monitor


For Legionella specifically, HSE states that results should be interpreted in the context of the water system and the existing risk assessment, with subsequent action determined accordingly.
